Getting ready to swap pumps. Do the water input and output fittings require a gasket? What stops them from leaking? And is there a gasket/seal where it bolts to the engine?

Inlet/outlet flanges have o-rings. Pump base to timing gear housing has a gasket. All can be re-used if in good shape, but best to go new.
 
That particular pump has a poor reputation based on the quality of its internal components. What engine is it on? If a Cummins there is likely a compelling alternative available from Seaboard in California
 
+! on the alternative from Seaboard. Also no need for a key with the impeller and the impeller comes with a removal tool.
 
If you are leaving the hoses on the in/out fittings, pay particular attention in keeping the o rings in place while you snug things up. The hoses tend to hold them askew sometimes, if the o ring gets pinched it will look ok but leak.
